Abstract:

PROBLEM TO BE SOLVED: To obtain a new DNA synthase capable of efficiently amplifying the whole length of a template DNA strand, by artificially modifying an amino acid sequence so that the synthetic DNA stand extend longer in catalyzing the synthesis of the DNA strand complementary to a single-stranded DNA.
SOLUTION: This new DNA synthase which has an amino acid sequence of the formula or the like, is obtained by artificially modifying the amino acid sequence of PfuDNA synthase so that the synthetic DNA strand extend longer in catalyzing the synthesis of a DNA strand complementary to a single-stranded DNA. When a high-molecular DNA is to be amplified by PCR or the like, this new DNA synthase enables the whole length of the template DNA to be synthesized and amplified in high efficiency, thus enables the base sequence thereof to be easily determined in high accuracy. Specifically, this new DNA synthase is obtained by the following procedures: a genomic DNA is first extracted from the microbial cells of Pyrococcus friosus DSM 3638, and then cloned, through PCR technique, with a primer synthesized according to the base sequence of PfuDNA synthase gene, and the resulting gene is integrated into a vector and then expressed in host cells.
