Abstract:

To provide a fed-batch fermentation process which uses special E. coli host/vector systems for the purpose of efficiently forming recombinant proteins, in particular recombinant antibody molecules, preferably antibody fragments such as miniantibodies.

An E. coli expression vector which is suitable for expressing exogenous proteins under high cell density fermentation conditions is constituted by using at least (i) an upstream terminator sequence and a downstream terminator sequence, (ii) lac promoter/operator system, (iii) a T7g10 Shine/Dalgarno sequence, (iv) pelB or ompA signal sequence, and (v) the sequence of an exogenous gene.
