Abstract:
To provide alternative and robust systems and techniques for sequence targeting with a wide array of applications.SOLUTION: Provided is a CRISPR-Cas vector system comprising one or more vectors comprising: a) a first regulatory element operably linked to a nucleotide sequence encoding a Cas9 protein containing one or more nuclear localization signaling (NLS); b) a second regulatory element operably linked to a nucleotide sequence encoding a CRISPR-Cas guide RNA comprising a guide sequence that can hybridize to a target sequence in eukaryotic cells and can direct sequence-specific binding of a CRISPR-Cas complex to the target sequence, a tracr mate sequence that can hybridize to a tracr sequence, and a tracr sequence; and c) a recombinant template, the components (a), (b), and (c) being located on the same or different vectors of the system.SELECTED DRAWING: None
