Abstract:

PURPOSE: To perform rapid and highly accurate immunoassay using a small amount of a specimen, by performing quantification based on a sandwich method using a monoclonal antibody to human III, IV or VI type collagen peptide.

CONSTITUTION: After an antibody-containing substance is fractionated using ammonium sulfate as an antibody to which an enzyme marker is applied, an IgG fraction purified by a DEAE-Sephacel column is used and, in the case of a polyclonal antibody, a substance purified by a Sepharose 4β affinity column coupled with human III, IV or VI type collagen is used. Further, as a monoclonal antibody and a polyclonal antibody, the specific bonding parts F(ab')2 or Fab' in said antibodies may be used. By measuring the concn. of human III, IV or VI type collagen peptide by using the monoclonal antibody to said collagen in one of or both of the antibody bonded to a solid phase carrier and the antibody to which the enzyme marker is applied, the forecasting of a trouble in the liver is achieved without performing biopsy being a heavy burden on a patient.
