Abstract:

PURPOSE: To obtain an enantiomer of a heterodicyclic alcohol by stereoselectively resolving from its corresponding racemic body through a specific process having no need of an optically active reagent, while converting another enantiomer into the racemic body.

CONSTITUTION: The racemic alcohol of the formula (X is O, S or the like; Y1-Y3 are independently H, a halogen, a 1-4C alkyl; NO2 bonds to the bicyclic ring system at the 5- or 7-position; C* atom has R or S configuration) is acylated with an acylating agent under influence of an enzyme that has a stereoselective esterification activity. Unesterified compounds are removed from the ester to isolate the substantially pure alcohol enantiomer (ester) represented by the formula. Then, the ester is hydrolyzed to convert it to the corresponding alcohol enantiomer, while the undesired enantiomer is converted into the starting racemic enantiomer under basic conditions.
