Abstract:

PURPOSE: To efficiently convert preproinsulin into insulin useful for treatment, etc., of diabetes by hydrolyzing the preproinsulin in the presence of clostripain and, as necessary, treating the resultant product with carboxypeptidase B.

CONSTITUTION: A preproinsulin represented by the formula [R1 is (n) x amino acids and (n) is 0 or 1; R2 is H, an amino acid which can be cleaved off chemically or enzymatically or a peptide having 2-30 amino acid residues; R3 is hydroxyl, an amino acid or a peptide having 2-10 amino acids; X is L-arginine or a peptide with 2-45 amino acids and with a C-terminal and N-terminal L- arginine residue; Y and Z are each a genetically encodable amino acid; A1 to A20 or B2 to B29 is an amino acid squence which is natural or has been modified by replacement of one or more amino acid residues of human or animal insulin] is hydrolyzed in the presence of clostripain and, as necessary, converted with carboxypeptidase B into the corresponding insulin.
