Abstract:

PURPOSE: To obtain Escherichia coli carrying a plasmid forming tryptophan comprising an Escherichia strain having an autoamplifying cloning vector and a desensitized tryptophan operon and capable of efficiently producing L- tryptophan by culturing the Escherichia stain.

CONSTITUTION: This Escherichia coli carrying a plasmid forming tryptophan is obtained by isolating tryptophan operon (trpI15) from Escherichia coli W3110 trpEA1trpRtnaA (PSC101trpT15), connecting the operon to autoamplifying cloning vector pTK235 isolated from Escherichia coli C600, introducing the connected vector to Escherichia coli EA2 (Escherichia coli WB3110tnaA2 trpEA2) strain to provide a transformed Escherichia coli WB30trpEA2tnaA (pKTtrp20) ATCC67399. L-Tryptophan is obtained by culturing the strain in a nutrient medium at pH about T at 30-40°C and isolating the L-tryptophan from the obtained medium.
