To obtain the subject composition enabling estrogen therapy and the supplement of estrogen to mammals, in which the increase of prolactin level accompanying estrogen administration is unwanted, by compounding dehydroestorone, etc., having neutralizing effect on prolactin level as an active component.

This composition contains 8,9-dehydroestorone or its 3-sulfuric ester, which is pharmaceutically permissible. A daily dose of this composition is preferably 0.02μg/kg to 500μg/kg in terms of the active compound. This administration is useful for the treatment of a state of disease or symptom in which estrogen administration is beneficial but the increase of prolactin level is unwanted, for example, infertility, or ovulation disorder in amenorrhea, etc., or osteopenia, which is often accompanied by hyperprolactinemia.
