Abstract:

PURPOSE: To obtain a plasmid for expression in Escherichia coli, having a high hydroxylating activity, capable of expressing cytochrome P-450MC having wide substrate specificity in the Escherichia coli, and applicable to oxidation reaction processes on an industrial level and oxidative removal of organic compounds in industrial waste water, etc.

CONSTITUTION: A plasmid pMG1 obtained by linking a structural gene of cytochrome P-450MC isolated from a recombinant plasmid pAU157 (producible by the method described in Nud. Acids. Res. 12, P2929W2938) having the whole coding region of rat hepatic cytochrome P-450MC protein to a tac promoter which is a powerful promoter of Escherichia coli. The tac promoter is constituted of -35 region of trp promoter and -10 region of lacUV5 promoter.
