Abstract:

NEW MATERIAL:A protein bonding to the Fc fragment of immunoglobulin G. It has the following properties. Molecular weight, 24,000; isoelectric point, 4.5pH; ultraviolet absorption, absorption peak at 275nm (E1=1.60); bonding capacity to human immunoglobulin G, 11mg/mg.

USE: Useful as an agent for the purification and concentration of antibody or as a clinical diagnostic agent. It specifically bonds to the Fc fragment of immunoglobulin G and the bonding capacity per unit weight is superior to that of protein A.

PREPARATION: Staphylococcus aureus SR-1 (deposited as a deposition No.B-1964 in Central Research Lab., Sanraku Co. Ltd.) is cultured at 37°C for 24hr e.g. by shaking culture and the objective compound is separated from the cultured liquid. The Staphylococcus aureus SR-1 is a mutant of Staphylococcus aureus Cowan I (ATCC 12598).
