To obtain the subject fertile plant capable of stably expressing a heritable recombinant DNA integrated into a chromosome by setting regenerable and fertile callus cultures from a plant Zea mays to be transformed and treating the resultant transformed cells under specific conditions.
Callus cultures such as a friable embryo callus culture initiated from a regenerable and fertile immature hybrid embryo are set from a plant Zea mays to be transformed and the set cell cultures are then bombarded with recombinant DNA-coated microprojectiles such as a bacterium dap A gene to thereby transform the cultured cells. The transformed cells are subsequently identified or selected to thereby regenerate a fertile transgenic plant Zea mays capable of stably expressing a heritable recombinant DNA. As a result, the transgenic plant Zea mays is obtained. The callus cultures to be bombarded are preferably groups of 30-80 mg per group.
