To provide a fluorescence reader for acquiring a fluorescence detection image with low back noise even if a gel cassette having intense self-fluorescence is used.
In a fluorescence detection image acquired by a CCD camera 3 through an optical filter, even self-fluorescence components of the gel cassette 7 can appear in addition to a specimen fluorescence component of a nucleic acid in gel 1 for migration. Therefore, in addition to the specimen fluorescence components of the nucleic acid in the gel 1 for migration and the self-fluorescence components of the gel cassette 7, a first comparative image in which even a reflected light component of the gel cassette 7 appears is acquired by the CCD camera 3 not through any optical filter. By using a luminance ratio between pixels corresponding to a specific position of the gel cassette 7 where only the gel cassette 7 appears, a second comparative image is prepared by modifying the brightness of the first comparative image so that it is even with the brightness of the detection image to perform inter-pixel subtraction on the second comparative image with respect to the detection image. A corrected image is thus acquired into which the detection image is picture-processed.
