Abstract:
PROBLEM TO BE SOLVED: To provide a method using a genetically modified phage that does not restore its lytic property when the phase is used to produce a biomolecule, thereby avoiding a culture medium from being contaminated with an infectious phage or preventing the phase from being inadvertently dissolved during growth or protein production.SOLUTION: According to the present invention, there is provided a genetically modified phase in which, based on the fact that ral gene and N gene are related to a lysogenic state and deletion of a DNA fragment containing coding sequences of ral gene and N gene leads to the death of the host cell, at least one gene selected from Int gene, Xis gene, R gene, S gene, Q gene, and Rz gene is inactivated. There is also provided a process for producing a biomolecule using a host cell containing the genetically modified phage. The genetically modified phase comprises a T7 expression system as an expression system and is defined as a lambda phage, 434 phage, phi80 phage, phi81 phage, HK97 phage, or P21 phage, or one from P 11 phage to P 53 phage.SELECTED DRAWING: Figure 1
