Abstract:
PURPOSE: To obtain highly stable, heat-resistant adenosine-5'-phosphosulfate kinase by culture of Bacillus bacteria and by taking an enzyme having specific physicochemical characteristics from the cultured product.
CONSTITUTION: Bacillus bacteria such as Bacillus coagulans are aerobically cultured in a nutrient medium pref. at 60°C for 2 to 6hr, and the resulting microbial cells are collected, crushed by e.g. ultrasonic treatment, put to nucleic acid removal, and purified by e.g. chromatography, thus obtaining the objective enzyme. This enzyme has the following physicochemical characteristics: (1) activity: forming 3'-phosphoadenosine-5'-phosphosulfate by acting on adenosine-5'- triphosphate and adenosine-5'-phosphosulfate; (2) optimal pH: 7.5-9.5; temperature for activity: 50-70°C; heat resistance: activity after treated in a buffer solution at ca.60°C for ca.15min retains 95% of that before treatment; and (3) molecular weight: ca. 50000 (determined by gel filtration).
