Abstract:
PROBLEM TO BE SOLVED: To obtain the subject new enzyme consisting of a fused protein of a fructosylamino acid oxidase having a specific amino acid sequence with a glutathione S-transferase, excellent in thermal stability, and useful in clinical and food fields. SOLUTION: This new heat resistant fructosylamino acid oxidase is a fused protein of a polypeptide having an amino acid sequence described in the formula or an amino acid sequence in which one or several amino acids are deleted, substituted or added, with a glutasthione S-transferase, consists of a recombinant protein having a fructosylamino acid oxidase activity, is useful for the analysis of an Amadori compound in clinical and food fields, and used for the index of the symptom of diabetes mellitus and the quality control of the food. The enzyme is obtained by culturing a transformed body obtained by incorporating a DNA encoding the fused protein of the fructosylamino acid oxidase with glutathione S-transferase.
