Abstract:
To increase the expression amount of a useful protein containing a disulfide bond which is difficult to obtain high productivity by using a recombinant Escherichia coli, especially an antibody such as scFv by rapidly transferring the protein into the periplasm in order to improve the productivity of the antibody, or to provide a novel expression method for increasing the expression amount (display amount) on the outer membrane surface of Escherichia coli using a system for displaying as a fusion protein with an outer membrane protein.SOLUTION: When a useful protein such as scFv containing a disulfide bond is produced by using a recombinant Escherichia coli, a "cell permeability-improving peptide tag" consisting of any one of five amino acid residues selected from a specific amino acid sequence is linked immediately after the C-terminal of a periplasmic transfer signal.SELECTED DRAWING: None
