PROBLEM TO BE SOLVED: To improve the efficiency of ligating single stranded DNAs together.SOLUTION: By adding several bases of ribonucleotide using terminal deoxynucleotidyl transferase (TdT) to an acceptor side single stranded DNA beforehand, the efficiency of a ligation reaction of the 3' end of the acceptor side single stranded DNA to the donor side single stranded DNA, the 5' end of which has been phosphorylated beforehand, by using RNA ligase is enhanced.SELECTED DRAWING: None
