PURPOSE: To obtain the subject deriv. for an enzyme transporting agent for transfusion, etc., which persists in blood vessels and has a low oxygen affinity by covalent coupling the tetrameter of the hemoglobin and a polycarboxyl compd. under conditions under which both are crosslinked between dimers.
CONSTITUTION: An aq. soln. of the polycarboxyl compd. (e.g. benzene hexacarboxylate, etc.), having 4 to 6 calboxyl groups in the molecule is added to a soln. of the tetrameter of hemoglobin molecules, such as oxyhemoglobin and the pH of the soln. mixture is adjusted to 6.6 by using 0.1 M HCl. An N'-ethyl-N-(3-dimethylaminopropy)carbodiimide chlorohydrate is then added to the soln. under stirring to cause the covalent coupling under the conditions under which a crosslinking is formed between the two dimers of the hemoglobin molecules. The objective human hemoglobin deriv. having such oxygen affinity to allow an in-vivo to use of the bond in the state that the natural allosteric effector of hemoglobin does not exist is thereby obtd.
