PURPOSE: To obtain an easily handleable and soluble human thrombomodulin derivative comprising a specific amino acid sequence containing the N-terminal, epidermal growth factor homology and serine/threonine-rich regions of the human thrombomodulin, capable of being utilized for the treatment and prevention of thrombus.
CONSTITUTION: This amino acid sequence comprises, in order from the N- terminus, (a) the N-terminal, (b) epidermal growth factor homology, and (c) serine/threonine-rich regions of human thrombomodulin. The amino acid sequence lacks the transmembrane and cytoplasmic domains of the human thrombomodulin, and can be derived from AV 12 or 293 host cell transformed with a recombinant DNA vector encoding the amino acid sequence. The amino acid sequence of the soluble thrombomodulin derivative is preferably an amino acid sequence encoded with the thromobodulin derivative-encoding sequence of plasmid pUC18TMD1 or plasmid phdTMD1.
