To provide a hyperthermophilic duplex-specific nuclease (DSN) and a method for cleaving nucleic acid using it.
Provided are a protein derived from Coenobitidae having duplex-specific nuclease activity, a gene encoding the protein, a recombinant vector containing the gene, and a transformant or transductant including the vector. Also provided is a method for obtaining a protein having duplex-specific nuclease activity characterized by culturing the transformant or transductant in a culture medium, and collecting the protein having duplex-specific nuclease activity from the culture. Also provided are a method for cleaving nucleic acid by using the protein having duplex-specific nuclease activity, and a method for detecting RNA using the protein, and a reagent kit containing the protein.
