PURPOSE: To provide a protein produced with a specific human ovarian tumor cell, having a specific mol.wt., exhibiting a multiplication-stimulating activity for vascular endothelial cells, not exhibiting the multiplication-stimulating activity for vascular smooth muscular cells, etc., and useful for cure-stimulating agent for wounds, burns, etc.
CONSTITUTION: HVOCA II (FERN 2310) and HVOCA III (FERM 2311) which are human ovarian tumor cells or human ovarian tumor-established cells are cultured in a serum-free culturing medium. The supernatant of the serum-free culturing product is separated, passed through a cation exchange column, and eluted with a NaCl linear gradient. An active fraction is passed through an immobilized heparin column, and an active fraction is eluted with a NaCl linear gradient to provide the objective human-originated single strand protein having a SDS-PAGE mol.wt. of 72000-80000 dalton under a non-reducing condition, a SDS-PAGE mol.wt. of 79000-85000 under a reducing condition, having a multiplication-stimulating activity for vascular endothelial cells, not having the multiplication-stimulating activity for fibroblasts, vascular smooth muscle cells, and hepatic cells, and having a vascularizing activity.
