Abstract:
2-(Allenyl)penicillins antibiotics and intermediates thereto are disclosed. The antibiotics have the formula oror a mixture of the two, wherein:R1 is hydrogen or a methyl group;R2 is hydrogen, an amino-protecting group or a C1 to C20 acyl group; andRz is a carboxy-protecting group or hydrogen; or a salt thereof; which comprises preparing a compound when R2 is a group of the formulawherein R6 is a group of the formulaand Rz is hydrogen, or a salt thereof, by combining a substrate compound of the formulaor a substrate compound of the formulawith the enzyme known as isopenicillin N synthetase, wherein in the above formulasR1 is hydrogen or methyl; andR2a is a group of the formulaorwherein R23 and R24 are hydrogen, and optionally converting the product to a compound in which R2 is hydrogen, another acyl group or an amino-protecting group.
