To provide a method for immobilizing a compound, capable of immobilizing the compound such as a ligand for specific adsorption in a high yield and capable of highly maintaining the activity of an immobilized SH group- containing compound by reacting a solvent-insoluble carrier with an SH group- containing compound in the presence of an antioxidizing agent.
This method for immobilizing a compound comprises reacting an SH group-containing compound (for example, cystein) with a carrier activated with at least one kind of functional group selected from glycidyl group, imidocarbonate group, tosyl group, tresyl group, carboxyl group, amino group, azido group, hydroxyl group, etc., as a solvent-insoluble carrier (for example, agarose) in the presence of at least one antioxidizing agent selected from sodium pyrosulfite, sodium disulfite, sodium sulfite, sodium bisulfite, sodium hydrosulfite, L-ascorbic acid, etc. Thus, the SH(sulfhydryl) group-containing compound is immobilized on the solvent-insoluble carrier in a high yield.
