PURPOSE: To establish the immunological measuring method of lentinan by combining a certain kind of protein with lentinan to synthesize a lentinan-protein conjugate, and immunizing an animal with the conjugate as an antigen to obtaining anti-lentinan antiserum for immunological measurements.
CONSTITUTION: Sodium periodate and lentinan are brought into reaction for oxidative cleavage. The resulting aldehyde group is combined with protein to produce a lentinan-protein conjugate with a Schiff base. This conjugate is used as an antigen, and the animal is immunized. Thus, the antiserum which reacts with the lentinan is obtained. When the antiserum which reacts with the lentinan is obtained. When the lentinan and the animal serum are incubated, the antiserum is added. Thus, the complement activating reaction of the lentinan is neutralized with the antiserum. Then, the antiserum is used, the specific immunoreaction for the lentinan is utilized, and the concentration of the lentinan is measured.
