Abstract:

PURPOSE: To detect an antibody or an antigen by use of a solid particle on which an antigen or an antibody is fixed on the surface by chemical bond and by observation of a physical variation caused by change of a electric change of said particle before or after antigen-antibody reaction.

CONSTITUTION: After adding 5g polymer particles of 0.174μm average particle size contg. 0.922meq/g carboxy group to water of 4pH adjusted by hydrochloric acid, 0.5g 1-cyclohexyl-3-(2-morpholinoethyl)carbodiimid used for a ordinary peptide synthesis and 4ml of 1,8-diamino-4-aminomethyl octane are added. The mixture is stirred and reacted at room temperature for 2hr and followed by stirring for 1hr in a aqueous solution of a 25ml phosphate buffer solution of 7.7pH and 25ml glutar aldehyde. After 10ml a phosphate buffer solution of 7.6pH contg. 0.3mg an antihuman serum albumin is added and stirred for 1hr, an antihuman serum albumin fixed to solid particles is produced. These particles and a human serum albumin is mixed and stirred, and electrophoresis is performed and velocity of transfer is determined by a microscope.
