Abstract:
PURPOSE: To prepare choline oxidase, in high yield, by culturing a bacterial strain capable of producing choline oxidase, in a medium containing choline.
CONSTITUTION: A bacterial strain capable of producing choline oxidase, e.g. Arthrobacter globiformis B-0577 (FERM-P No.3518), is inoculated in a medium containing 0.5W1% choline based on the medium, and cultured by the aerated and agitated deep-tank cultivation techinque. The objective choline oxidase having substrate specificity to the compound of formula A (R1 is -CH2OH or -CHO) and exhibiting the enzymatic activity shown in the formulasIand/or II can be separated from the cultured bacterial cells.
