Abstract:

PURPOSE: To obtain a 17-ester useful as an anti-inflammatory agent in high yield, by hydrolyzing a 17α,21-cyclic orthoester of 17α-,21-dihydroxy-20-keto-steroid under specific conditions.

CONSTITUTION: A 17α,21-cyclic orthoester of a 17α,21-dihydroxy-20-keto-steroid having the 17-position and a side chain thereof in the D ring expressed by the partial structural formula (R is 1W10C alkyl, aralkyl, cycloalkyl or aryl; R' is 1W4C lower alkyl) is hydrolyzed in the presence of a Lewis acid, e.g. aluminum chloride, in an aqueous alcoholic medium to prepare 17-monoester of 17α,21-dihydroxy-20- keto-steroid. The formation of the 21-ester and the 17α,21-diol as by-products is very slight.
