Abstract:
PURPOSE: To economically obtain 7-aminoacyl-cepharosporin without using a halogen-contg. solvent by allowing a N-substd. vinyl α-aminoacid to react with an acylating agent and then allowing the product to react with a cephem- carboxylic acid.
CONSTITUTION: A N-substd. vinyl α-aminoacid is allowed to react with an acylating reagent (e.g. pivaloyl chloride) in a halogen-free solvent having no miscibility with water or having little solubility with water (e.g. methylisobutylketone) at a temp. of -50 to 50°C to obtain a mixture of carboxylic acid anhydride. Then the obtd. soln. is allowed to react with a 7- amino-ceph-3-em-4-carboxylic acid expressed by formula I (Y is O, S, methylene; R1 is H, halogen, alkoxy, etc.; R2 is H, cation, carboxy protecting group, etc.; R' is H, amino protecting group) in a halogen-free solvent (e.g. n-butyl acetate) to obtain a compd. expressed by formula II (R is phenyl, 4-hydroxyphenyl, 1,4-cyclohexadien-1-yl).
