Abstract:

PURPOSE: To obtain the subject inhibitor, capable of inhibiting a key enzyme guanosine diphospho-D-mannonse dehydrogenase for biosynthesis of alginates by Pseudomonas aeruginosa and useful as a therapeutic agent for intractable infectious diseases, especially diffuse panbronchiolitis due to the Pseudomonas aeruginosa.
CONSTITUTION: This inhibitor of bacterial biofilm formation contains a compound expressed by formula I (R1 is H, a 1-5C alkyl or an aglycone part of a 12- to a 16-membered ring macrolide; R2 is H or hydroxyl group) or its pharmacologically permissible salts as an active ingredient. 9- Dehydrodemycarosylplatenomycin, etc., in which R1 is the group expressed by formula II and R2 is hydroxyl group are cited as the compound expressed by formula I. The 9-dehydrodemycarosylplatenomycin can be prepared by cleaving mycarose obtained by acylating mydecamycin A3 which is a 16-membered ring macrolide. Furthermore, the dose of the compound expressed by formula I for an adult is about 0.01-30mg/kg, preferably 0.1-10mg/kg.
