Abstract:

PURPOSE: To provide a new ketone derivative having high inhibitory activity on thiol protease, also excellent in peroral absorbability, intertissue migration capability and permeability through cell membrane.
CONSTITUTION: The objective compound of formula I [R1 is H or formulas II to V (R10 is alkyl, aryl or heterocyclic residue); R2, R4 and R6 are each H or alkyl; R3 and R5 are each H, aralkyloxy, aryl, alkyl, etc.; R2 and R3 and R4 and R5 are respectively combined into N-contg. heterocycles; R7 is H, alkyl, aralkyloxy, aryl or alkoxy; R8 is H, alkyl or aralkyl; R7 and R8 are combined into benzylidene or cycloalkyl; A is S, SO, SO2, O or NR11 (R11 is H or alkyl); R9 is aryl or (CH2)m-X (X is H, OH, amino, halogen, aryl, etc.; m is 0-15) when A is other than 0, being H or (CH2)1-X (l is 1-15) when A is 0; n is 0 or 1], e.g. (S)-3-[(S)-2-amino-4-methylvalerylamino]-1-furfurylthio-2-heptanone.
