Abstract:
PROBLEM TO BE SOLVED: To provide a method for separating and concentrating leukocytes, capable of extremely preventing the contamination and staining of the erythrocytes, capable of strictly grasping the number of transplanted hematopoietic stem cells, and capable of being operated in an aseptic state. SOLUTION: This method for separating and concentrating leukocytes comprises slightly centrifuging or hanging a blood mother liquid collected in a mother bag 1 in a state that the attaching positions of child bags 3, 4 to the parent bag 1 are placed under the parent bag 1, transferring the precipitated erythrocytes in the lower layer into one child bag 3, separating the child bag 3 from the parent bag 1, slightly centrifuging the parent bag 1 in a state that the attaching position of the child bag 4 to the parent bag 1 is placed on the parent bag 1, thus separating the leukocyte layer into the upper plasma layer and the lower concentrated leukocyte solution layer, gradually transferring the upper plasma into the child bag 4, aseptically adding a prescribed amount of a frost damage-preventing agent into the parent bag 1 receiving the concentrated leukocyte solution, and subsequently transferring the concentrated leukocyte solution into a freezing bag.
