Abstract:
To provide a mutant of a protein and a DNA for the production of the mutant by searching for a protein bonding to Mec1 of a budding yeast belonging to the ATM relating factor.
A factor Pie1 interacting with Mec1 of budding yeast is separated, the function of check point control of the Pie1 is analyzed and the mutants of the protein pie1-KA and pie1-ΔC are prepared. The pie1-KA is a mutant produced by substituting the 177th and 178th lysines of Pie1 with alanines and pie1-ΔC is a mutant obtained by depleting 47 amino acids from the C-terminal of Pie1. The abnormality in the control of cell proliferation and check point response has been experimentally confirmed on these mutants.
