Abstract:

Prodn. of 2-(2-hydroxyethoxy)ethyl N-(alpha, alpha, alpha-trifluoro-m-tolyl)anthranilate (I) comprises first converting N-(alpha, alpha, alpha-trifluoro-m-tolyl)anthranilic acid (II) in presence of an inert organic solvent to its acid halide, then reacting this with 2-(2-hydroxyethoxy)ethanol (III) at 0-50 degrees C. Pref. first stage is at 0-30 degrees C using PCl5 as halogenating agent, and the second at 30-50 degrees C using a 7.5-15 times excess of (III). The pref. solvents are toluene and xylene and reaction is esp. carried out under inert gas. Process gives an almost quantitative yield of good purity prod. which may be further purified, without significant materials loss, by simple distn. Pharmaceutical-grade material can be made without need for chromatographic sepn.
