Abstract:
PROBLEM TO BE SOLVED: To provide efficient, cost-effective, and large-scale methods of manufacturing carrier polypeptide.SOLUTION: The invention relates to methods comprising incorporating a first unnatural amino acid into a carrier polypeptide variant, incorporating a second unnatural amino acid into a target polypeptide variant, reacting the first and second unnatural amino acids to produce the conjugate, and further making a covalently coupled carrier polypeptide-target polypeptide conjugate. The invention also relates to orthogonal translation systems in methylotrophic yeast and to methods of producing carrier and target polypeptide variants containing unnatural amino acids using these systems.SELECTED DRAWING: None
