Title:
MANUFACTURE OF NITROGEN-CONTAINING TETRACYCLIC COMPOUND
Document Type and Number:
Japanese Patent JPS56135487
Kind Code:
A
Abstract:
1388582 Quinobenzoxazepine- and quinobenzothiazepine-derived amines E R SQUIBB & SONS Inc 5 June 1972 [24 June 1971] 26100/72 Heading C2C The invention comprises compounds of formulµ and their salts, wherein X, Y are each C 1-8 alkyl, alkoxy or alkylmercapto, C 3-7 cycloalkyl, halogen, CF 3 , -CN, -NC or di(C 1-8 alkyl)- sulphamyl; A is O or S; R1, R2 are each H, C 1-8 alkyl, aralkyl or C 2-8 alkenyl, or NR1R2 is wherein X1 is NR4, O, S or CH 2 , r is 1-3 and R3, R4 are each H or C 1-8 alkyl [possibly substituted by OH, C 1-8 alkanoyloxy or hydroxyalkoxy, NH 2 (possibly substituted by 1 or 2 C 1-8 alkyl groups) or di(C 1-8 alkyl)amino-C 1-8 - alkoxy]; and n, n1 are each 0-2. These compounds may be prepared by reacting the 3-oxo analogues with hydroxylamine or HNR1R2 and reducing the oximes or enamines so formed, or by reacting the 3-chloro analogues (obtained from the 3-hydroxy compounds) with HNR1R2. Alternatively when NR1R2 is a heterocyclic ring, the latter may be formed by reacting with an open-chain dihalide, the compound where R1= R2=H. Starting materials.-Details are given for the preparation of the appropriately substituted 3- (5-dibenzo-oxazepine and -thiazepine) propionic acids, nitriles, methyl esters and acid chlorides. Pharmaceutical compositions which may be administered orally or parenterally as CNS stimulants, muscle relaxants or anti-inflammatory agents; veterinary compositions which may be administered orally or topically as antimicrobial agents; or surface disinfectants in the form of a dust, spray, soap or other detergent composition, comprise compounds of the above formula.
