PURPOSE: To readily and accurately measure a microbial cell concentration in a reaction solution, by adding a protein solubilizing agent to a solution sampled from a reaction vessel to dissolve microbial cells, removing suspended materials and measuring the fluorescent intensity of the resultant solution.
CONSTITUTION: A solution is partially sampled from a reaction vessel for microorganisms. A protein solubilizing agent is then added to the sampled solution to dissolve the microorganisms. The resultant solution is further subjected to solid-liquid separation treatment to remove suspended materials and give a transparent solution, which is then irradiated with excitation light at a given wavelength and the intensity of fluorescence emitted by the protein components in the solution is measured. The protein solubilizing agent is preferably SDS (sodium dodecyl sulfate) and the amount thereof is preferably 2W20 times based on the expected microbial cell concentration. The solution is preferably heated at 80W100°C for 5W10min in dissolving the microorganisms. The excitation light has 260W320nm wavelength and the wavelength of the fluorescence for measurement is preferably 320W380nm.
