To provide a novel quantifying method of pyrophosphoric acid in a sample, a novel measuring method of nucleic acid synthesizing reaction using it and a system for these methods.
In a system for detecting the concentration of pyrophosphoric acid by using an ion responsive field effect transistor (ISFET) sensor, a sensor provided with a tantalum pentoxide layer is designated as the ISFET sensor. Especially, in the measuring method of the concentration of pyrophosphoric acid in the sample, the cumulative type ISFET sensor, which is constituted so as to measure the charge quantity accumulated after potential is reset as a potential change, corresponding to the depth of a potential well changed, corresponding to the concentration of the ions acting on a sensing part and the pump-out number of times from the potential well is used. The measuring method of nucleic acid synthesizing reaction is constituted so as to measure the change in the concentration of pyrophosphoric acid which is produced as the byproduct of the polymerase reaction to target nucleic acid, by using the measuring method of the concentration of pyrophosphoric acid. At least the ISFET sensor and the system for measuring the nucleic acid synthesizing reaction in the sample having polymerase activity are constituted.
