To provide a method for introducing a biologically active peptide into a host by administering a polynucleotide which operatively encodes the peptide.
The polynucleotide which is naked in the meaning that it is neither complexed with a colloidal preparation such as a liposome or associated with a genomic integrating vector, is administered. In one embodiment, the naked polynucleotide is administered to a host tissue which has a high concentration of antigen presenting cells when compared with other host tissues. It is believed that the polynucleotide is taken in and expressed locally, systemically and/or at locations remote from its intake point in the host. In another embodiment, the polynucleotide express a peptide which is effective in retarding, treating and preventing age-associated diseases and delays loss due disease resistance in mammals. In this embodiment, the peptide of a lower level than a therapeutic level of the peptide is expressed at an appropriate stage of host growth. Devices and compositions for use in the methods of this invention are also described.
