To obtain the subject composition excellent in inhibitory action on proliferation of mesangium cell, effective for an animal nephritis model, low in toxicity, useful for treating nephritis, containing a specific compound as an active ingredient.
This composition comprises a hederagenin derivative of the formula {Me is CH3; R1 is an OR11 or an NR11R12 [R11 and R12 are each H, a (substituted) alkyl, a (substituted) cycloalkyl, etc.]; R2 and R3 are each H, a (substituted) alkyl, a (substituted) cycloalkyl, etc.} or its pharmaceutically permissible salt or a solvate of any thereof such as hederagenin (3β,23- dihydroxyolean-12-en-28-oic acid as an active ingredient. The compound, for example, is obtained by extracting Spindus mukurossi in a raw state as it is in a hydrous alcohol solvent at 50-80°C for 1-4 hours, concentrating the extracted solution under reduced pressure, hydrolyzing the concentrated solution with a solution of a mineral acid in an alcohol solvent, adjusting the solution to pH 6-7, adding activated carbon and purifying under heating.
