Abstract:

PURPOSE: A messenger RNA enabling the production of human interleukin 2 by gene recombination operation, obtained by extracting and separating it from a cell derived from human lymphocyte.

CONSTITUTION: A cell derived from human lymphocyte capable of producing spontaneously human interleukin 2 or producing it by bringing it into contact with a mitogen in the presence or absence of another cell so that it is stimulated is cultivated using a medium for common cell cultivation until the production of interleukin 2 starts in the supernatant liquid of the culture. The prepared cell is partially or completely destroyed, solubilized and mRNA is extracted. mRNA corresponding to the desired human interleukin is obtained as 11W12S fraction by fractionation through density gradient centrifugation method of sugar. The corresponding DNA is prepared from the mRNA, and human interleukin 2 can be prepared by manifesting this DNA in a cell of a microorganism.
