PURPOSE: To decrease the concentration of salt in a moving phase and to make it possible to perform detection so as not to shorten the life of an apparatus by using a material wherein specified quantities of tetrabutyl ammonium and organic solvent are mixed to the specified quantity of buffer at a specified concentration as the moving phase.
CONSTITUTION: In a moving-phase tank 3, the following material is contained as a moving phase: a material wherein 2-30-millimol tetrabutyl ammonium and 5-30% organic solvent are added in phosphoric acid buffer solution whose hydrogen ion concentration is adjusted at pH 3-8. Transition metal ions are added into a specimen solution to be analyzed, and the specimen which is transformed into the chelate of iron-ethylenediaminetetraacetic acid is prepared. When the specimen is injected into a specimen injecting mechanism 2, the chelate is subjected to the action of the organic solvent in the moving phase and the tetrabutyl ammonium. The specimen is separated in a reverse-phase distributing column 1 which is filled with a filler whose functional group is octadecyl group. Light absorption occurs, and a sharp peak is generated. Ultraviolet-ray absorbance is measured 4 based on the peak.
