Abstract:
PROBLEM TO BE SOLVED: To provide a method for multiplex polymerase chain reaction (PCR) amplification of short tandem repeat (STR) loci that can be used to rapidly generate a highly specific STR profile from target nucleic acids.SOLUTION: According to the present invention, there is provided a method for detecting the presence of single nucleotide polymorphisms (SNPs) of a nucleic acid in a sample containing the nucleic acid. The method comprises: contacting in a solution the sample with eight or more unlabeled oligonucleotides under conditions permitting hybridization between the oligonucleotides and the nucleic acids in the sample; generating primer extension products; in the presence of a plurality of ddNTPs each labeled with one of eight or more different fluorescent dyes, labeling each product with the fluorescent dye by a primer extension reaction; and detecting the SNPs by detecting the products of the primer extension, each product labeled with the fluorescent dye by laser-induced fluorescence using a galvanometer in a step-and-stare mode and a spectrograph to detect the fluorescence from each of the fluorescent dyes.SELECTED DRAWING: None
