Abstract:

PURPOSE: To efficiently obtain purified cell bodies of a Clostridium microorganism by culturing the microorganism in an aqueous alkaline medium whose pH is kept within a certain range, promoting the proliferation of cell bodies, inhibiting the bacteriolysis of the cell bodies, and densifying the culturing of the microorganism.

CONSTITUTION: A bacterium of Clostridium, for example, Clostridium butyricum FERM BP-2789 strain is inoculated to a culture medium so that the initial concentration of cell bodies becomes 106 cells/ml and cultured at 37°C under mild stirring of 20rpm at a pH ranging from 4.5-6.5 using a pH controller of an aqueous alkali such as 5% aqueous NaOH solution, until the cell body density reaches the maximum value, as the pH is kept constant. Thus, the proliferation of the bacterium is promoted, as their bacteriolysis is inhibited to efficiently give pure Clostridium cell bodies which are useful as an antitumor agent, and immunoactivating agent, an antibody-enhancing preparation, therapeutic agent for animals including human, food products and fertilizer.
