Abstract:
PURPOSE: To extract an antigen originating from all of serotypes of bacteria while lowering a background by extracting an antigen originating from bacteria accompanied by a periodontal disease by using a buffered extraction compsn. to mix the same with a blocking compsn. and bringing the resulting mixture into contact with an antibody specific to the antigen to form an immune complex.
CONSTITUTION: An antigen originating from bacteria accompanied by a periodontal disease is extracted by using an extraction compsn. at least bufferred to pH8 and the extract compsn. is mixed with a blocking compsn. containing a sufficient amt. of nonimmune blocking protein to form a mixture containing at least 0.2wt.% of protein and not receiving the adverse effect of high pH or a present surfactant because of the blocking properties of protein. The mixture is brought into contact with an antibody specific to the antigen without lowering the pH of the mixture to 8 or less to form an immune complex which is, in turn, detected. A diagnostic examination kit contains an extract bufferred to pH8, a water-soluble antibody specific to bacteria accompanied by a periodontal disease and nonimmune blocking protein.
