Abstract:

PURPOSE: To develop an extraneous polypeptide at a level substantially higher than the master strain by platting the cell of fungi developing different kind of polypeptide on a solid medium and bonding the cell of fungi with an antibody through selective mutual reaction.

CONSTITUTION: Master strain of fungi of Saccharomyces cerevisiae is sprayed or applied using a wire onto a solid medium composed of agarose and plated to develop a different kinds of polypeptide, e.g. human albumin, human PAI-2 or human α1-antitrypsin. They are then bonded selectively with an antibody and a ring of precipitated antibody/polypeptide is observed visually as it is or after dyeing and labelling. Consequently, an extraneous polypeptide can be developed at a level higher than the master strain and the observation is facilitated.
