Abstract:

To provide a method for detecting a target base sequence by which the bonding affinity between a nucleic acid aptamer and a ligand is heightened, and the target base sequence in a specimen can be detected even if the concentration is low; and to provide a reagent for enhancing detection sensitivity, and a kit for the detection.

The method for detecting the target base sequence in the test specimen comprises (a) a step for forming a nucleic acid aptamer obtained by hybridizing the target base sequence in the test specimen with a probe by mixing the test specimen with the probe hybridizable with the target base sequence, (b) a step for reacting the nucleic acid aptamer with the ligand in the presence of a compound having ≥1.8 dipole moment in the reaction solution, and (c) a step for detecting the target base sequence by using the bonding affinity of the nucleic acid aptamer with the ligand as an index.
