PURPOSE: To simply and accurately measure the subject enzyme by mixing a substrate carrier and an activation carrier with a specimen solution to color- develop the solution, the substrate carrier containing 2-chloro-4-nitrophenyl-α- maltotrioside, etc., as a substrate and the activation carrier containing an azide compound as an activation agent.
CONSTITUTION: A substrate carrier 2 and an activation carrier 3 are successively put on a support 1 such as a polystyrene plate so as to contact with each other or laminated to each other. A specimen soltion such as human urine is dropped on the carriers to color-develop the solution, and the developed color is measured with a densitometer to determined amylase. The substrate carrier 2 is produced by impregnating filter paper with an aqueous solution containing 2-chloro-4-nitrophenyl-αmaltotrioside or 4-nitrophenyl-α-maltotrioside as a substrate, NaCl, calcium acetate and a surfactant, drying the paper and cutting the dried paper. The activation carrier 3 is also produced by impregnating filter paper with an aqueous solution containing 0.1-5% of an azide compound such as sodium azide, drying the filter paper and subsequently cutting the dried filter paper.
