Abstract:

PURPOSE: To accurately determine a thermal history of milk or the number of elapsed days after production thereof.
CONSTITUTION: A coloring liquid containing sodium 3'-{1-[phenyl amino- carbonyl]-3, 4-tetrazolium-bis(4-methoxy-6-nitro)benzenesulfonate is mixed with milk to be reacted then a coloring degree is measured. 2-methyl-1,4- naphthoquinone are added to the coloring liquid as mediator. The measuring the the coloring degree is executed measuring variations of an absorbance at a wavelength of 492nm, or measuring variations of a color tone before and after coloring reaction by utilizing a color-difference meter as variations of a value (a). The evaluation kit of milk is constituted of a coloring reagent containing sodium 3'-{1-[phenylamino-carbonyl]-3,4-tetrazolium}-bis(4-methoxy-6- nitro)benzenesolfonate and 2-methyl-1,4-naphthoquinone, a micro plate, and a micro pipette.
