Abstract:
PROBLEM TO BE SOLVED: To provide a technique allowing for suppression of evaporation or air bubbles generation in a liquid sample in a microchip during heating, with a simpler constitution.SOLUTION: A microchip 1 has a substrate 2, a protective member 3, a reaction vessel (sealed space) 5, and a gas generating film (gas generating agent) 6. The gas generating film 6 is comprised of a photoresponsive or thermal responsive gas generating agent. In heating a liquid sample 10 housed in the reaction vessel 5, external stimulation, such as light, is given to the gas generating film 6 to generate gas. Thus, pressure by gas is applied to the reaction vessel 5, and a liquid sample 10 can be heated while pressure is applied thereto. A heating method of the present invention can be used in a degeneration (dissociation) process or the like in nucleic acid amplification reaction.
