Abstract:
PURPOSE: To stably keep a recombinant plasmid and to stably produce the objective useful substance in high productivity by culturing a yeast holding a recombinant plasmid in a medium added with histidine.
CONSTITUTION: A yeast holding a recombinant plasmid is cultured in a medium containing ≥150mg/l of histidine preferably at 20-35°C for 2-4 days. The recombinant plasmid is preferably plasmid pGEL125, etc., for the extracellular manifestation of human lysozyme and the host is e.g. Saccharomyces cerevisiae AH22R-/pGLD P31-RcT (IFO 10226).
